No. 1: Batman & Superman

Despite completely different upbringings the two are without a doubt one of the most well known and greatest bromances in comic book history. Superman's amazing foster parents Johnathan and Martha Kent raising Clark in Smallville, Kansas to be great man and use his godlike powers humbly to save people. And Batman losing his parents Thomas and Martha Wayne at gunpoint changing him forever and pushing him to become the caped crusader seeking justice for his parents murder. While they live in cities that the other wouldn't be caught dead in, these two help each other out even in the most trying of times and trust each other more than anyone. They've also revealed their secret identities to each other on purpose or not (Superman has used his x-ray vision to see under the cowl, and Batman just figured it out).
In stories like Public Enemies where Lex Luthor becomes president and forces heroes to sign up under his government (much like Marvel's Civil War) these two friends stand by each other fighting old allies and enemies saving each other's lives many times throughout the story many times. Or even starting the Justice League together along with other friends and allies knowing that just the two of them can't do it alone. These two also have had their differences through stories like The Dark Knight Rises where they are on opposite sides of the law and even the recently released Batman v Superman: Dawn of Justice though they soon became allies.
